The hospital was founded on April 3, 1876 by Dr. Daniel Pérez Márquez and Dr. Francisco de Paula Pérez Márquez, two doctors who were born in San Juan de la Maguana and studied medicine at the University of Santo Domingo.
San Juan de la Maguana Hospital has been operating since 1876 and has undergone many changes over the years, including its location from San Juan to La Vega in 1948, its name change to “Hospital General Gregorio Luperón” in 1958, and finally becoming “Hospital General San Juan de la Maguana” on January 1st, 2006 as a result of a merger with another hospital in La Vega called “Hospital Santiago”.
